ABSTRACT

Objective:To explore the clinical effects of intramuscular injection of oryzanol for 5 and 10 days in the treatment of menopausal syndrome.Methods:This study adopts a case-control study method.Twenty patients with menopausal syndrome admitted to Peking Union Medical College Hospital from July 2021 to March 2022 were selected and randomly divided into a control group and an observation group using a random number table method, with 10 patients in each group. The control group was given intramuscular injection of oryzanol once a day for 5 consecutive days on the basis of routine health education; On the basis of routine health education, the observation group was given intramuscular injection of oryzanol once a day for 10 consecutive days. Two groups of patients completed a total of 6 visits before treatment and on days 5, 10, 30, 60, and 90 of treatment. Baseline survey was conducted to record the basic information of patients, including demography data, menstruation, past history, surgical history, drug allergy and the main symptoms of climacteric syndrome, and the baseline hormone level of patients was detected. Compare the menopausal symptoms by modified K-score, Pittsburgh sleep quality index (PSQI) score, menopausal specific quality of life (MENQOL) score, post treatment injection site pain by visual analog score (VAS), and observe adverse reactions during treatment between the two groups of patients at each visit time point.The measurement data conforming to the normal distribution is expressed in xˉ± s. The comparison between the two groups is performed by t test, and the change trend of the two groups at each time point is compared by repeated measurement analysis of variance; The measurement data that do not conform to the normal distribution are represented by M( Q1, Q3), and the Kruskal Wallis test is used for comparison between groups; Counting data is represented by examples(%), Fisher's exact probability method is used for inter group comparison, and Wilcoxon rank sum test is used for rank data comparison. Results:The body mass of the control group was higher than that of the observation group [(62.50±11.88) kg vs (52.90±4.28) kg, t=2.40, P=0.027]. There was no statistical difference between the two groups in other demography data and previous history, sex hormone levels, menopausal symptoms, and menstrual conditions (all P>0.05). Compared with the baseline, the modified K-score, PSQI score, and MENQOL vascular dilation and contraction, psychological state, physiological state, and sexual life scores of the two groups of patients on the 5th, 10th, 30th, 60th, and 90th day of treatment were significantly reduced, and showed a gradually decreasing trend with the extension of observation time (the values in Group F were 36.37, 12.40, 18.69, 24.73, 40.77, and 18.39, respectively, P<0.001), but there was no statistically significant difference between the groups (all P>0.05). After treatment, the VAS score of the control group was 1 point in 2 cases, 2 points in 4 cases, and 3 points in 4 cases; The VAS score of the observation group was 1 in 4 cases, 2 in 3 cases, and 3, 5, and 6 in 1 case each. There was no statistically significant difference in VAS scores between the two groups ( Z=0.43, P=0.664), and no adverse events or serious adverse events were observed during the treatment period. Conclusions:Intramuscular injection of oryzanol injection for 5 and 10 days can improve menopausal symptoms, improve sleep quality and overall quality of life in patients with menopausal syndrome, and the overall effect is similar.

ABSTRACT

Objective: The objectives of this study were to quantify γ-oryzanol in an ethanolic extract of Oryza sativa L. Indica (black rice) bran and to evaluate its activity as a tyrosinase inhibitor. Methods: Black rice bran was extracted via maceration in 96% ethanol, and the γ-oryzanol concentration in the extract was measured through high-performance liquid chromatography. The applicability of the extract as a skin lightening agent was determined by evaluating its tyrosinase inhibition activity. Results: The dry rice bran contained 118.572 mg/g of γ-oryzanol, and the extract inhibited tyrosinase activity at an IC50 of 74.8%. Conclusion: The black rice bran extract was sufficiently potent for use in skin lightening formulations.

@#Introduction: Rice bran oil (RBO) is used in Asian countries as a daily dietary supplement. RBO is known in particular for its hypolipidemic effect. There has been increasing interest recently in the use of RBO as a means to maintain body weight and prevent obesity, though the mechanism of how this happens is still not well understood. We have investigated the effect of RBO on expression of genes that might influence energy homeostasis and feed intake. Methods: This study assessed Sprague-Dawley male rats at 12-weeks that were split into three groups over a 28-day period. A control group was fed a diet of standard rat chow, a standard group was fed standard rat chow with Orlistat (10.8 mg/kg bw/day), and a treatment group was fed standard rat chow with RBO (57.6 mg oryzanol/day). All supplementation was given by oral gavage. Possible adiposity was investigated through a histological analysis of adipocytes size measurement of intra-abdominal white adipose tissue in the rats. Changes in gene expression in the liver were determined by microarray. Results: The data suggest that RBO supplementation of a regular diet did not result in excess body weight and adiposity. A microarray analysis of the rats’ livers found that RBO altered the expression of genes related to energy homeostasis and feeding behavior, by upregulating genes such as Olr522, RGD1561231 and Rgs16. Conclusion: It is suggested that RBO supplementation can be used to maintain body weight by lowering appetite.

ABSTRACT

Objective@#To evaluate the clinical efficacy of auricular acupoint application in the treatment of burning mouth syndrome(BMS).@*Methods@#A total of 155 patients diagnosed with BMS were randomly divided into the auricular acupoint application group (50 patients), drug treatment group (55 patients), and auricular acupoint application combined with drug treatment group (50 patients). One month represented one course of treatment. The changes in pain intensity were evaluated before treatment as well as one month and three months after treatment.@* Results@#The VAS scores in the auricular acupoint application group (t=8.949), the drug treatment group (t=10.52) and the auricular acupoint application combined with drug treatment group (t=19.33) all decreased 1 month after treatment, with a statistically significant difference compared with the scores before treatment (P < 0.01). The VAS scores of the auricular acupoint application combined with drug treatment group decreased significantly, and the difference was statistically significant compared with the scores in the drug treatment group (t=3.91, P=0.000 2). 3 months after treatment, the VAS scores of the three group decreased compared with that before treatment, but increased compared with that 1 month after treatment, and the VAS score of the drug treatment group increased most obviously, but the difference was not statistically significant compared with that of the auricular acupoint application group (t=2.047, P=0.043), other pairwise comparison differences were not statistically significant. There was no statistically significant difference in VAS score in the auricular acupoint application group (t=1.752) and in the drug treatment group (t=0.174) compared with that before treatment (P > 0.05). Compared with before treatment, the VAS score in the auricular acupoint application combined with drug treatment group also decreased significantly (t=3.282, P < 0.05). @*Conclusion@#Auricular point application is a safe and effective treatment for burning mouth syndrome, and the long-term effect is better when combined with drugs.

ABSTRACT

This study investigated the effect of ionic liquid (IL)-microwave assisted extraction (MAE) on γ-oryzanol extraction from rice bran (Oryza sativa L.). The conditions included the concentration of IL, liquid (IL)/solid (sample) ratio, extraction time, and microwave power. The use of 1-butyl-3-methylimidazolium ([Bmim]BF4) as a selected solvent in IL-MAE method was developed for extraction of γ-oryzanol from rice bran. The Box-Behnken design on four factors with response surface methodology (RSM) was used to optimize the experimental conditions. The optimum conditions for the MAE were a [Bmim]BF4 concentration of 0.7 M, liquid/solid ratio of 15 mL/g, extraction time of 10 minutes, and 30% microwave power with a γ-oryzanol value of 0.41 mg/g. The experimental values agreed with those predicted by the RSM models. A microplate reader method was used to determine the IC50 value in tyrosinase inhibitory activity of the extract. An IC50 value of the extract obtained from IL-MAE was 1240 µg/mL. This study suggests that IL-MAE is suitable for the extraction of γ-oryzanol from rice bran and that under the optimum conditions the extract has low activity as a tyrosinase inhibitor.

ABSTRACT

Rice bran (RB) is one of the nutrient-rich agricultural byproducts. It is a composite of carbohydrates, lipids, proteins, fibers, minerals, and trace elements such as phosphorus, potassium, magnesium, calcium and manganese. The extraction and purification process influences the quality and quantity of rice bran oil, which is rich in tocopherols, tocotrienols, γ-oryzanol, and unsaturated fatty acids. The bioactive components of RB have been reported for exhibiting antioxidant, anti-inflammatory, hypocholesterolemic, anti-cancer, anti-colitis, and antidiabetic properties. In vitro and in vivo studies, and clinical trials in human volunteers revealed the anti-hyperglycemic activity of RB derived compounds. An updated comprehensive review on the antidiabetic property of RB and its derivative is required to appraise the current knowledge in the particular field. Thus, the present paper covered the composition and bioactivities of RB, and influence of extraction methods on the biological property of rice bran oil and rice bran extract. And the current review also focused on the reported anti-hyperglycemia activity of rice bran derivatives, and its probable mechanism.

ABSTRACT

ObjectiveTo explore clinical effect and safety ofShaoyao-Gancao decoction combined withSiwu decoction for the patients with diabetic peripheral neuropathy (DPN).MethodsA total of 76 patients with DPN in our hospital from 2013 July to October 2015 were divided into control group and observation group by the random number method, 38 patients in each group. The control group were treated with conventional treatment combined with oryzanol injection, and the observation group were treated with Shaoyao-Gancaodecoction combined withSiwu decoction on the basis of control group. Both groups were treated for one month. The level of FPG, 2 hPG, HbAlc, TC, TG, LDL-C and HDL-C was detected respectively before and after treatment. The whole blood viscosity and plasma viscosity was detected with LBY-N6A self-cleaning rotary viscosimeter. The red blood cells deposited and platelet aggregation rate were tested with Winchester points vascular method and turbidimetric method, respectively.The leg nerve and sural nerve of nerve conduction velocity (nerve conduction velocity, NCV) were determined with electromyography, at the same time the clinical curative effect was evaluted.ResultsTotal effect rate of observation group was 89.5% (34/38), which was significantly higher than the control group 71.1% (27/38), and the difference was statistically significant (χ2=4.070,P=0.044); After treatment, the levels of FPG (6.12 ± 0.38 mmol/Lvs.6.58 ± 0.52 mmol/L, t=4.403), 2 hPG (7.83 ± 0.82 mmol/Lvs. 8.41 ± 0.93 mmol/L,t=2.884), HbAlc (6.27 ± 0.52%vs. 6.82 ± 0.64%, t=4.112) in the observation group were lower than those in the control group (P<0.05). The levels of TC (4.73 ± 0.83 mmol/L vs. 5.11 ± 0.64 mmol/L,t=52.235), TG (1.83 ± 0.35 mmol/Lvs. 2.03 ± 0.41 mmol/L, t=2.287) and LDL-C (2.91 ± 0.54 mmol/Lvs. 3.25 ± 0.58 mmol/L,t=2.645) in the observation group were lower than those in the control group (P<0.05), and the HDL-C (1.47 ± 0.33 mmol/Lvs. 1.26 ± 0.31 mmol/L,t=-2.859) in the observation group was higher than than that in the control group(P<0.05). The level of whole blood specific viscosity (10.16 ± 2.12 mPa?svs. 11.33 ± 2.51 mPa?s,t=2.195), plasma viscosity (1.24 ± 0.25 mPa?svs. 1.62 ± 0.37 mPa?s,t=5.246), red blood cell volume (0.31 ± 0.16L/Lvs. 0.42 ± 0.08 L/L,t=3.791) and platelet aggregation rate (50.21% ± 8.03%vs. 54.16% ± 7.82%,t=2.172) in the observation group were significantly lower than those in control group (P<0.05). The tibial nerve velocity (45.22 ± 3.20 m/svs. 38.26 ± 5.19 m/s,t=-7.037) and the common nerve velocity (43.22 ± 6.34 m/svs. 36.23 ± 4.81,t=-5.415) in the observation group were as significantly higher than those in control group (P<0.05).ConclusionsThe Shaoyao-Gancaodecoction combined withSiwu decoction could improve the state of blood sugar, blood lipid and blood rheology of patients with DPN, and improve nerve transfer speed of leg nerve and sural nerve, the curative effect is better than conventional western medicine therapy alone.

Objective To study Curative efficacy of Oryzanol combine with Dingkundan in treatment of perimenopausal period and its effects on endocrine improvement.Methods 90 patients of perimenopausal period who received therapy from March 2014 to March 2015 in our hospital were selected as research objects.According to draw method divided into the experimental group(n=45)and the control group(n=45).Two groups of patients were psychological support.The control group was treated with Oryzanol, each 10 mg, 3 times a day.while the experimental group was on the control group, treated with Dingkundan, 1 pills each time, 2 times a day, in the morning and evening.Then Serum estradiol(E2), Follicle stimulating hormone(FSH), Luteinizing hormone(LH)level, upset hot sweating, insomnia, irritability, hung up improve time, Kupperman score, clinical efficacy of two groups were compared.Results The E2 level in experimental group was significantly higher than the control group[(72.51±5.30)pg/mL vs.(66.80±4.75)pg/mL](P<0.05), FSH、LH level was significantly lower than the control group[(21.36±1.05)mIU/ml vs.(26.09±1.89)mIU/mL,(24.34±1.01)mIU/mL vs.(20.61±1.50)mIU/mL](P<0.05);upset hot sweating, insomnia, irritability, hung up improve time was significantly lower than the control group[(11.21±2.35)d vs.(20.05±5.07)d,(12.78±2.09)d vs.(18.32±4.13)d,(11.30±2.10)d vs.(20.16±5.02)d,(10.20±2.76)d vs.(16.58±4.03)d](P<0.05);Kupperman score was significantly lower than the control group[(4.32±1.09)score vs.(12.76±3.16)score](P<0.05);total effective rate was significantly higher than the control group[93.33%(42/45)vs.75.67%(34/45)](P<0.05).Conclusion Oryzanol combine with Dingkundan is well for perimenopausal period,which can significantly improve endocrine disorders, promote its balance, improve estrogen.

El propósito de esta investigación fue determinar el contenido de algunos minerales, de ácido fítico y de γ-oryzanol en harinas integrales y salvados de tres cultivares (Cimarrón, Fonaiap 1 y Zeta 15) de arroz (Oryza sativa L) cosechados en Venezuela. Los minerales (Ca, Mg, Fe y Zn) fueron cuantificados por espectrofotometría de absorción atómica, mientras que el fósforo (P) y el ácido fítico fueron estimados a través de un método colorimétrico. El γ-oryzanol se cuantificó por HPLC. En general, el contenido de minerales fue mayor en la harina y en el salvado de la variedad Zeta 15. El contenido de P (400,0 ± 5,00 mg/100 g) y Mg (60,0 ± 0,00 mg/100 g) fue menor en la harina de la variedad Cimarrón, mientras que en los salvados de las distintas variedades se observaron valores significativamente altos de P (1860 ± 2,00 - 2185 ± 0,58 mg/100 g) y Ca (66,7 ± 1,15 ­ 73,3 ± 1,53 mg/100 g). Las tres variedades evaluadas representan importantes fuentes de ácido fítico y γ-oryzanol; sin embargo, la variedad Cimarrón fue la que exhibió las menores concentraciones de estos compuestos bioactivos. Basado en los resultados obtenidos, las harinas integrales y los salvados representan una fuente importante de minerales, de ácido fítico y de γ-oryzanol. La concentración de todos los microcomponentes es significativamente superior en los salvados con respecto a las harinas, lo que los convierte en una atractiva alternativa para la obtención de compuestos con actividad biológica de interés para la salud humana(AU)


The objective of this study was to determine the content of some minerals, phytic acid and γ-oryzanol in whole-grain flour and bran in three cultivars (Cimarrón, Fonaiap 1 y Zeta 15) of rice (Oryza sativa L) in Venezuela. The minerals were (Ca, Mg, Fe y Zn) quantified by Atomic Absorption Spectrophotometry while that the phosphorus (P) and phytic acid were determined using a colorimetric method. The γ-oryzanol was quantified by HPLC. In general, the mineral contents were higher in flour than bran in the cultivar Zeta 15. The P (400,0 ± 5,00 mg/100 g) and Mg (60,0 ± 0,00 mg/100 g) contents were lower in flour of the cultivar Cimarrón, while that were observed higher values of P (1860 ± 2,00 - 2185 ± 0,58 mg/100 g) and Ca (66,7 ± 1,15 ­ 73,3 ± 1,53 mg/100 g) in the bran of different cultivars. The three cultivars evaluated are an important source of phytic acid and γ-oryzanol; however, the Cimarrón cultivar exhibited the lowest concentrations of these bioactive compounds. Based on these results, whole-grain flours and brans may offer a good source of minerals, phytic acid and γ-oryzanol. The concentrations of all micro compounds evaluated were higher in the brans than the flours, this become the bran an attractive alternative to obtain compounds with biological activity of interest for human health(AU)

ABSTRACT

Background & objectives: Enhanced muscle strength is seen when resistance exercise is combined with the consumption of nutritional supplements. Although there is a limited number of studies available about the efficacy of gamma oryzanol supplementation with resistance exercise in humans, but its usage as a nutritional supplement for strength is common in athletes. The aim of this study was to determine the effects of gamma oryzanol supplementation during 9-week resistance training on muscular strength and anthropometric measurements of young healthy males. Methods: In this double-blind clinical trial, changes of anthropometric measurements and muscular strength were studied after chronic resistance exercise and gamma oryzanol supplementation in 30 healthy volunteers (16 in supplement and 14 in placebo). Each day, gamma oryzanol supplement (600 mg) and placebo (the same amount of lactose) were consumed after training. The participants exercised with 80 per cent 1-Repetition Maximum (1-RM), for one hour and four days/week. Anthropometric measurements and subjects’ 1-RM for muscular strength were determined at the commencement and end of the 9-week study. Results: There was no significant difference between the baseline characteristics and target variables at baseline between the two groups. After gamma oryzanol supplementation, there was no significant difference in the means of anthropometric and skin fold measurements between the supplement and placebo groups. However, there were significant differences between the supplement and placebo groups for 1-RM of bench press and leg curl, which showed that gamma oryzanol improved muscle strength following resistance training. Interpretation & conclusions: Our findings indicated that 600 mg/day gamma oryzanol supplementation during the 9-week resistance training did not change anthropometric and body measurements, but it increased muscular strength in young healthy males. Further, studies need to be done in trained athletes, women, and in patients who suffer from muscular fatigue.

Drought-tolerant cultivars and their phytochemical composition, which has a role in providing drought tolerance are gaining importance. In this study, rice bran oil and semi-purified oryzanol (SPO) obtained from five rice (Oryza sativa L.) cultivars, namely P1401 and PB1 (drought-susceptible) and N22, PNR381 and APO (drought-tolerant) were analyzed for the γ-oryzanol content, an antioxidant present in considerable amount in the rice bran. The higher level of γ-oryzanol and its antioxidant activity was observed in drought-tolerant cultivars (N22, PNR381 and APO) as compared to drought-susceptible (PB1 and P1401), suggesting the role of γ-oryzanol in drought tolerance, as antioxidants are known to play an important role by scavenging free radicals. The total antioxidant activity of γ-oryzanol might be attributed to 24-methylene cycloartanyl ferulate, a major component of γ-oryzanol. By enhancing the level of active oryzanol components identified in this study by genetic and molecular means could impart increased drought tolerance.

The results obtained by these two different quantification methods were compared by paired t-test.Results:Both assays provided good linearity, accuracy, reproducibility and selectivity for determination of γ-oryzanol. Conclusions: The TLC-densitometric and TLC-image analysis methods provided a similar reproducibility, accuracy and selectivity for the quantitative determination of γ-oryzanol in cold pressed rice bran oil. A statistical comparison of the quantitative determinations of γ-oryzanol in samples did not show any statistically significant difference between TLC-densitometric and TLC-image analysis methods. As both methods were found to be equal, they therefore can be used for the determination of γ-oryzanol in cold pressed rice bran oil.

Several studies have implicated the involvement of poor glycemic control and oxidative/nitrosative stress in the development of diabetic neuropathic pain, an important microvascular complication affecting more than 50% of diabetic patients. However, lack of understanding of the underlying etiology, development of tolerance, inadequate relief and possible toxicity associated with classical analgesics warrant the investigation of the novel agents. Therefore, the present study was carried out to investigate the effect of oryzanol (OZ), a commercially-important potent antioxidant component isolated from from crude rice bran oil (cRBO), in streptozotocin (STZ)-induced diabetic neuropathy in rats. After eight weeks, diabetic rats developed neuropathy which was evident from decreased tail-flick latency (thermal hyperalgesia) and increased nociceptive behavior during the formalin test. This was accompanied by decreased motor coordination based on the evaluation of neuromuscular strength. Na+ K+ ATPase, a biochemical marker associated with the development of diabetic neuropathy, was significantly inhibited in the sciatic nerve of diabetic animals. The activities of antioxidant enzymes and lipid peroxidation levels were significantly elevated in diabetic rats, indicating the involvement of oxidative stress in diabetic neuropathy. Chronic treatment with oryzanol (OZ) (50 and 100 mg/kg) per oral (p.o.) and standard drug glibenclamide (Gl) (10 mg/kg, p.o.) significantly attenuated the behavioral as well as biochemical changes associated with diabetic neuropathy. The findings provide experimental evidence to the protective effects of OZ on hyperglycemia-induced thermal hyperalgesia and oxidative stress which might be responsible for diabetes induced nerve damage.

Experimental studies carried out for evaluating the anti-hyperlipidemic properties of rice bran components have given interesting but often contrasting results. Therefore, the current study was initiated to investigate the anti-hyperlipidemic activity of oryzanol (OZ), a commercially-important bioactive phytochemical, isolated from crude rice bran oil (cRBO). OZ was isolated by a two-step solvent crystallization process from cRBO, which was extracted from fresh rice bran by hexane mediated soxhlet extraction. Subsequently, OZ (50 and 100 mg/kg, p.o.) was evaluated for anti-hyperlipidemic activity in Triton WR-1339-induced acute hyperlipidemic albino rats by estimating serum triacylglyceride (TG), total cholesterol (TC), very low density lipoprotein-cholesterol (VLDL-C), low-density lipoprotein-cholesterol (LDL-C) and high-density lipoprotein-cholesterol (HDL-C) levels with atorvastatin as the reference standard. The degree of protection was also assessed by measuring the levels of various hepatic anti-oxidant enzymes. OZ evoked a significant decrease in the levels of serum cholesterol, triacylglycerides, LDL, VLDL and a significant increase in the level of serum HDL and hepatic anti-oxidant enzymes. It also showed a significant ameliorative action on elevated atherogenic index (AI) and LDL/HDL-C ratios. These findings indicate that OZ possesses the potential to lower plasma lipid concentrations and might be of therapeutic benefit in hyperlipidemia and atherosclerosis.

ABSTRACT

This study was carried out to investigate the supplementary effects of gamma-oryzanol extracted from rice bran on lipid metabolism in diabetic mice. We supplied 2 kinds of experimental diets (CO without and GO with 0.2% gamma-oryzanol) to diabetic mice for 8 weeks. Diet intake, body weight, organ weight, contents of serum and hepatic lipid profiles, and fecal lipid levels were measured. Though there was no significant difference in diet intake, body weight change and organ weight between experimental groups, the concentration of serum total cholesterol and hepatic total lipid, total cholesterol and HMG-CoA reductase activity was significantly lower in GO group treated with 0.2% gamma-oryzanol of diet than CO group after supplementary period of experimental diets. And total lipid, triglyceride, total cholesterol, and bile acid levels excreted to feces were significantly higher in GO group than CO group. These results suggest that gamma-oryzanol decrease the serum and hepatic lipid levels by lowing HMG-CoA reductase activity or increasing the contents of fecal lipid in diabetic KK mice.
